将结核杆菌ESAT-6 和MPT-64 的编码基因分别插入真核表达载体JW 4303 构建重组质粒,用重组质粒对4 周龄的BALB/C和F1 代小鼠进行肌肉和皮内免疫,于末次免疫后10 天采血并分离血清,进行ELISA 检测。结果表明,小鼠能产生较强的体液免疫,特异性IgG 平均水平分别为参考血清的2.20 倍和3.19 倍,且对两种品系小鼠进行的相同途径的免疫,其抗体水平差异不显著;而不同途径的重组质粒免疫,抗体水平略有差异。
The coding genes of Mycobacterium tuberculosis ESAT-6 and MPT-64 were respectively inserted into the eukaryotic expression vector JW 4303 to construct the recombinant plasmids. The 4-week-old BALB / c and F1 mice were immunized intramuscularly and intracutaneously with the recombinant plasmids. Blood was collected 10 days after the immunization and the serum was separated for ELISA test. The results showed that the mice can produce strong humoral immunity, the average level of specific IgG were 2.20 times and 3.19 times that of the reference serum, respectively, and the immunization of the two strains of mice by the same route, the antibody level The difference was not significant; and different ways of recombinant plasmid immunity, antibody levels slightly different.
