目的:寻找一种简便有效测定壳寡糖的方法。方法:采用Elson-Morgan法、苯酚-硫酸法和3,5-二硝基水杨酸(DNS)法测定壳寡糖含量并进行比较。结果:Elson-Morgan法不适用于壳寡糖含量的测定;苯酚-硫酸法中盐酸氨基葡萄糖在200~3000μg范围与吸光度也有很好的线性关系,但苯酚-硫酸法对壳寡糖的测定值较实际值偏大;DNS法测定壳寡糖与实际值接近,平均回收率为96.1%,RSD为2.9%(n=6)。结论:DNS法操作简单、快速,重现性好,可直接用于壳寡糖含量的测定。
Objective: To find a simple and effective method for the determination of chitooligosaccharides. Methods: The content of chitooligosaccharides was determined by Elson-Morgan method, phenol-sulfuric acid method and 3,5-dinitrosalicylic acid (DNS) method and compared. Results: The Elson-Morgan method was not suitable for the determination of chitooligosaccharides. In the phenol-sulfuric acid method, there was a good linear relationship between the concentration of glucosamine hydrochloride and absorbance in the range of 200-3000 μg. However, the phenol- Which was larger than the actual value. The determination of chitooligosaccharides by the DNS method was close to the actual value with an average recovery of 96.1% and RSD of 2.9% (n = 6). Conclusion: The DNS method is simple, rapid and reproducible, and can be directly applied to the determination of chitooligosaccharides content.
