从箬叶中分离纯化得到两种箬叶多糖,采用氯磺酸吡啶作为硫酸酯化试剂,研究反应温度、时间及反应物的摩尔比对硫酸酯化过程的影响。得到产率80%以上,硫含量最高达219%的硫酸酯化多糖。发现硫酸酯化会使多糖构象发生一定的改变,硫酸酯多糖在特性粘度、比旋光度、园二色谱等性质方面有不同程度的变化。体外抗艾滋病病毒(HIV)实验表明硫酸酯化箬叶多糖比箬叶多糖具有更高的抑制HIV引起的MT4细胞病变性的作用,其最低有效浓度达5~10μg·ml-1,细胞毒性浓度为25mg·ml-1。
Two kinds of polysaccharides from leaves were isolated and purified from the leaves, and the effects of reaction temperature, reaction time and the molar ratio of the reactants on the sulfation process were studied by using chlorosulfonic acid-pyridine as the sulfation reagent. A sulfated polysaccharide with a yield of over 80% and a sulfur content of up to 21.9% was obtained. It was found that sulfation could change the conformation of polysaccharides. Sulfate polysaccharides changed to some extent in the aspects of intrinsic viscosity, specific rotation, and circular dichroism. In vitro anti-HIV test showed that sulfated polysaccharide had higher inhibitory effect on MT-4 cell disease caused by HIV than polysaccharides, the lowest effective concentration was 5 ~ 10μg · ml-1, Toxicity of 2 5mg · ml-1.
