目的研究卒中后抑郁大鼠脑内的成纤维细胞生长因子(FGF-2)的表达,探讨FGF-2与卒中后抑郁的关系。方法采用线栓大脑中动脉(MCAO)法制作卒中SD大鼠模型,将成模后的大鼠随机分为实验组和对照组。实验组采用孤养联合CMS来制作卒中后抑郁组,对照组为卒中大鼠,采用Western印迹法检测实验组与对照组大鼠额叶、海马组织中FGF-2蛋白表达水平,采用实时定量RT-PCR技术,检测两组间大鼠额叶组织中fgf-2 mRNA表达水平。结果实验组大鼠的额叶组织中FGF-2蛋白和fgf-2mRNA表达水平显著低于对照组(P<0.01),海马组织中FGF-2蛋白表达差异不显著(P>0.05)。结论卒中后抑郁大鼠额叶FGF-2表达水平下降是引起卒中后抑郁的原因之一。
Objective To investigate the expression of fibroblast growth factor (FGF-2) in the brain of post-stroke depression rats and to explore the relationship between FGF-2 and post-stroke depression. Methods Stroke SD rat model was established by MCAO. The rats were randomly divided into experimental group and control group. In the experimental group, the lonephary combined with CMS was used to make post-stroke depression group and the control group was stroke rats. Western blotting was used to detect the expression of FGF-2 in frontal lobe and hippocampus of experimental group and control group. Real- -PCR technique was used to detect the expression of fgf-2 mRNA in the frontal lobe of rats. Results The expression of FGF-2 and fgf-2 mRNA in the frontal lobe of the experimental group was significantly lower than that of the control group (P <0.01). The expression of FGF-2 in the hippocampus was not significantly different (P> 0.05). Conclusions The decreased expression of FGF-2 in the frontal lobe of post-stroke depression rats is one of the causes of post-stroke depression.
