目的:优选阳春砂多糖的超声提取工艺条件。方法:以阳春砂多糖提取率为评价指标,在单因素试验基础上,选定提取温度、时间及料液比为考察因素,通过三因素三水平Box-Behnken中心组合试验建立多糖提取率的二次多项式回归方程,经响应面回归分析得到优化组合条件。采用紫外分光光度法测定多糖含量。结果:最佳提取工艺条件为提取温度78℃,超声时间71 min,料液比1∶39,多糖提取率实测值(13.04±0.36)%,与预测值(13.22%)较为接近。结论:与传统水提取法相比,超声辅助法具有省时、高效的优点,为阳春砂多糖的后续产品研发提供实验依据。
OBJECTIVE: To optimize the ultrasonic extraction process conditions of Yangchun sand polysaccharide. Methods: Taking the extraction rate of the polysaccharide from the spring sand as the evaluation index, on the basis of the single factor experiment, selecting the extraction temperature, the time and the material-liquid ratio as the investigation factors, the three-factor and the three-level Box-Behnken center combination test established the polysaccharide extraction rate Sub-polynomial regression equation, the response surface regression analysis of the optimal combination conditions. Determination of polysaccharide content by UV spectrophotometry. Results: The optimal extraction conditions were: extraction temperature 78 ℃, ultrasonic time 71 min, ratio of material to liquid 1:39, extraction rate of polysaccharide (13.04 ± 0.36)%, which was close to the predicted value (13.22%). Conclusion: Compared with the traditional water extraction method, the ultrasonic assisted method has the advantages of saving time and high efficiency, and provides the experimental basis for the follow-up product development of the polysaccharide from the spring spring.
