目的:探讨阿司匹林作用下人子宫内膜腺癌Ishikawa细胞凋亡率与凋亡抑制蛋白Bcl-xl表达变化的关系。方法:应用流式细胞仪(FCM)检测不同浓度阿司匹林对子宫内膜腺癌Ishikawa细胞周期和凋亡率的影响;Westernblotting法检测阿司匹林对Bcl-xl蛋白表达的影响。结果:①阿司匹林引起细胞阻滞在G1期,细胞凋亡率增加,与药物作用浓度相关(P<0.05);②阿司匹林降低Bcl-xl蛋白的表达,与药物作用浓度相关(P<0.05);③Ishikawa细胞凋亡率与Bcl-xl蛋白的表达水平存在负相关关系(r=-0.873,P<0.05)。结论:Bcl-xl蛋白在阿司匹林促进子宫内膜腺癌细胞凋亡的调控中起到重要的作用。
Objective: To investigate the relationship between apoptosis rate of Ishikawa cells and the expression of Bcl-xl in human endometrial adenocarcinoma induced by aspirin. Methods: Flow cytometry (FCM) was used to detect the effect of different concentrations of aspirin on cell cycle and apoptosis rate of Ishikawa cells in endometrial adenocarcinoma. Western blotting was used to detect the effect of aspirin on Bcl-xl protein expression. Results: (1) Aspirin induced cell cycle arrest in G1 phase, and the apoptosis rate increased with the concentration of drug (P <0.05). ② Aspirin decreased the expression of Bcl-xl protein, which was related to the drug concentration (P <0.05). ③Ishikawa apoptosis rate and Bcl-xl protein expression level of negative correlation (r = -0.873, P <0.05). Conclusion: Bcl-xl protein plays an important role in the regulation of aspirin on apoptosis of endometrial adenocarcinoma cells.
