低氧诱导因子-1(HIF-1)是由两个亚基组成的异源二聚体体蛋白,其活性主要是由a亚基的稳定性来调节。它在低氧条件下被激活,特异性地与靶基因的低氧反应元件结合,并诱导靶基因转录,提高肿瘤细胞的低氧适应能力,利于其生长、增殖、转移。其机制包括促进肿瘤干细胞的分化,调节肿瘤细胞的代谢,促进肿瘤新生血管形成,抗肿瘤细胞凋亡等。近年有许多关于HIF-1与肝癌及肝癌细胞的研究,本文综述了其与肝癌发生、发展和治疗的研究现状。
Hypoxia-inducible factor-1 (HIF-1) is a heterodimeric body protein composed of two subunits whose activity is mainly regulated by the stability of a subunit. It is activated under hypoxic conditions, specifically with the target gene hypoxia response elements and induce target gene transcription, improve tumor cell hypoxia adaptability, and help its growth, proliferation and metastasis. Its mechanism includes promoting the differentiation of cancer stem cells, regulating the metabolism of tumor cells, promoting tumor angiogenesis, anti-tumor cell apoptosis and the like. In recent years, there are many researches about HIF-1 and hepatocellular carcinoma and hepatocellular carcinoma cells. This article reviews the research status of HIF-1 hepatocarcinogenesis, development and treatment.
