目的:探讨E-cadherin、β-catenin和PTEN在宫颈鳞癌及上皮内瘤变组织中的表达及临床意义。方法:采用免疫组化方法检测75例宫颈鳞癌、30例宫颈上皮内瘤变(CIN)及10例正常宫颈组织中E-cadherin、β-catenin和PTEN的表达。结果:正常宫颈、CINⅠ、CINⅡ~Ⅲ及宫颈鳞癌组织中E-cadherin阳性表达率依次为100.0%、85.7%、50.0%和37.3%;β-catenin异常表达率依次为0、28.6%、62.5%和84.0%;PTEN阳性表达率依次为100.0%、78.6%、56.3%和40.0%,3种蛋白在正常宫颈、CINⅠ的表达与鳞癌中的表达差异均有统计学意义,在正常宫颈与CINⅡ~Ⅲ中的表达差异也均有统计学意义,P<0.05。E-cadherin、PTEN在宫颈鳞癌中的阳性表达均与病理组织学分级、淋巴结转移及临床分期有关,P<0.05;β-catenin异常表达与病理组织学分级、淋巴结转移有关(P<0.05),但与临床分期无关(P>0.05)。宫颈鳞癌组织中E-cadherin与PTEN阳性表达呈正相关,与β-catenin异常表达呈负相关;β-catenin异常表达与PTEN阳性表达呈负相关。结论:E-cadherin与PTEN表达缺失或降低及β-catenin异常表达,均在宫颈鳞癌的发生、发展及淋巴结转移中发挥重要作用,且三者之间关系密切。
Objective: To investigate the expression and clinical significance of E-cadherin, β-catenin and PTEN in cervical squamous cell carcinoma and intraepithelial neoplasia. Methods: Immunohistochemistry was used to detect the expression of E-cadherin, β-catenin and PTEN in 75 cases of cervical squamous cell carcinoma, 30 cases of cervical intraepithelial neoplasia (CIN) and 10 cases of normal cervical tissue. Results: The positive expression rate of E-cadherin in normal cervix, CINⅠ, CINⅡ ~ Ⅲ and cervical squamous cell carcinoma was 100.0%, 85.7%, 50.0% and 37.3% respectively. The abnormal expression rates of β-catenin were 0,28.6%, 62.5 % And 84.0%, respectively. The positive expression rate of PTEN was 100.0%, 78.6%, 56.3% and 40.0% respectively. The expression of three proteins in normal cervix and CINⅠwas significantly different from that in squamous cell carcinoma. The expression difference in CINⅡ ~ Ⅲ also had statistical significance, P <0.05. The positive expression of E-cadherin and PTEN in cervical squamous cell carcinoma was related to histological grade, lymph node metastasis and clinical stage (P <0.05). Abnormal expression of β-catenin was associated with pathological grade and lymph node metastasis (P <0.05) , But not related to clinical stage (P> 0.05). Cervical squamous cell carcinoma E-cadherin positive correlation with PTEN expression, and β-catenin abnormal expression was negatively correlated; abnormal expression of β-catenin PTEN positive expression was negatively correlated. Conclusion: The loss or decrease of E-cadherin and PTEN expression and the abnormal expression of β-catenin all play an important role in the occurrence, development and lymph node metastasis of cervical squamous cell carcinoma, and the three are closely related.
