目的:为研究角膜上皮下混浊(haze) 形成的发生机理,检测激光角膜光学切除术(photorefractive keratectomy ,PRK) 后角膜上皮和基质表达转移生长因子- α(transforming growth factor- α,TGF-α) 的变化。方法:新西兰白兔16 只,随机分成4 组,其中12 只施行PRK。于术后1 、2 、3 个月用裂隙灯显微镜观察haze 形成情况,并用原位核酸分子杂交方法,检测角膜上皮和基质TGF- αm RNA 的表达。结果:PRK 后1 个月已有haze 形成,2 个月haze 最明显,术后3 个月haze 减轻或消失。正常角膜上皮有TGF- αm RNA 表达,基质中未见其表达。PRK 后角膜上皮表达TGF- αm RNA 增加,且以术后1 、2 个月表达最明显。TGF- αm RNA 表达强弱与形成haze 的轻重相关。结论:TGF- α参与PRK 后伤口愈合过程,且调节着haze 的形成和发展
OBJECTIVE: To investigate the mechanism of corneal haze formation and to detect the expression of transforming growth factor-α (TGF-β) in corneal epithelium and stroma after laser photorefractive keratectomy (PRK) α) changes. Methods: Sixteen New Zealand white rabbits were randomly divided into 4 groups, of which 12 were PRK. The haze formation was observed by slit lamp microscopy at 1, 2 and 3 months after operation, and the expression of TGF-αmRNA in corneal epithelium and stroma was detected by in situ hybridization. Results: One month after PRK, haze was formed, the haze was the most obvious in two months, haze was reduced or disappeared in three months after PRK. Normal corneal epithelium with TGF-αmRNA expression, no expression in the matrix. The expression of TGF-αmRNA in corneal epithelium after PRK increased, and the expression was most obvious at 1 and 2 months after surgery. The expression of TGF-αm RNA is related to the severity of haze. Conclusion: TGF-α participates in wound healing process after PRK and regulates the formation and development of haze
