通过共振散射光谱(RLS)、二维共振散射光谱(2D-RLS)及荧光光谱研究了牛血清白蛋白(BSA)与聚乙二醇(PEG)在溶液中的相互作用。PEG和BSA之间存在着氢键作用及疏水作用,其中氢键作用是主要的。在相同条件下,基于RLS方法测得的PEG与BSA之间的结合常数为9.56×103L/mol,而基于荧光光谱方法测得的PEG与BSA之间的结合常数为6.21×103L/mol。两种方法此外所测得的实验数据比较接近,表明RLS方法可普遍用来分析蛋白质与水溶性聚合物之间相互作用。
The interaction between bovine serum albumin (BSA) and polyethylene glycol (PEG) in solution was investigated by resonance scattering spectroscopy (RLS), two-dimensional resonance scattering spectroscopy (2D-RLS) and fluorescence spectroscopy. Hydrogen bonding and hydrophobic interaction exist between PEG and BSA, and hydrogen bonding is the main one. Under the same conditions, the binding constant between PEG and BSA based on the RLS method was 9.56 × 103 L / mol, while the binding constant between PEG and BSA based on the fluorescence spectroscopy method was 6.21 × 103 L / mol. In addition, the experimental data measured by the two methods are close to each other, indicating that the RLS method can be generally used to analyze the interaction between the protein and the water-soluble polymer.
