目的:建立反相高效液相色谱法测定干蟾皮药材中华蟾酥毒基的含量。方法:采用Phenomenex Gemini-C18色谱柱(250mm×4.6mm,5μm),流动相为乙腈-水(48∶52),流速0.8mL.min-1,检测波长296nm,柱温为35℃。结果:华蟾酥毒基浓度在19.6~174.6μg·mL-1范围内线性良好(r=0.9999);平均加样回收率(n=6)为99.9%,RSD为1.4%;不同批号的干蟾皮药材中华蟾酥毒基的含量为1.12%~1.16%。结论:本方法简便、准确,重复性好,适用于干蟾皮药材的质量控制研究。
Objective: To establish an RP-HPLC method for the determination of cinobufagin in dried toad skin. METHODS: Phenomenex Gemini-C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase was acetonitrile-water (48:52) with a flow rate of 0.8 mL.min-1. The detection wavelength was 296 nm and the column temperature was 35 ℃. Results: The concentration of cinobufagin was linear in the range of 19.6-174.6 μg · mL-1 (r = 0.9999). The average recovery was 99.9% (n = 6) and the RSD was 1.4% Chinese civet snake skin drug content of 1.12% to 1.16%. Conclusion: The method is simple, accurate and reproducible. It is suitable for the quality control of dried toad skin medicinal materials.
