目的从HA1基因层面探讨2007-2009年广西壮族自治区(广西)甲3流感病毒的变异、进化发展规律,探寻与WHO疫苗推荐株以及上海市、北京市和浙江省等地当年流行株之间的差异。方法提取广西22株2007-2009年甲3流感毒株RNA,并测定其血凝素重链(HA1)区域的核苷酸序列,采用生物信息软件,分析与当年疫苗推荐株及上海、北京和浙江等地在HAI以及HAI抗原决定簇区域的差异,分析进化关系。结果 22株广西甲3流感病毒2007年与2008年比较接近,HA1区核苷酸和氨基酸的同源性均值分别为99.39%和99.08%;而与2009年变异较大,同源性均值分别为99.04%和97.78%。而对比世界卫生组织(WHO)推荐的2008-2009年甲3流感疫苗株A/B risbane/10/2007,广西3年分离株HA1区氨基酸酸同源性均值分别为99.81%、99.25%和97.94%。2009年广西甲3流感病毒与当年疫苗株比较,HA1抗原决定簇发生了6个氨基酸的替换,且发生在A、D、E三区。各年度散发株和暴发株同源性较均在99.00%以上。2007年广西甲3毒株与上海及北京比,HA1区氨基酸同源性均值分别为99.83%和99.12%,2008年比较结果分别为98.93%和99.25%。基因进化树分析显示2009年分离株与2007年、2008年及当年疫苗株距离较远,单独分支。结论广西2007年与2008年甲3流感病毒HA1基因特性较为接近,2009年毒株发生了较大变异。广西与上海、浙江等地分离株更为接近,而与北京分离株差异较大。WHO推荐的当年疫苗株均滞后广西流行株。
Objective To investigate the variation and evolution of influenza A (A) virus in Guangxi Zhuang Autonomous Region (Guangxi) from 2007 to 2009 from the perspective of HA1 gene level and explore the relationship between the WHO vaccine recommended strains and the prevalence strains in Shanghai, Beijing and Zhejiang provinces difference. Methods The RNA of 22 strains of influenza A (H3PV) from 2007-2009 in Guangxi were extracted and sequenced. The nucleotide sequence of the hemagglutinin heavy chain (HA1) region was determined. Using bioinformatics software, Zhejiang and other places in the HAI and HAI antigenic determinants of the region differences, analysis of evolutionary relationships. Results 22 strains of GP-3 were close in 2007 and 2008 respectively. The average homology of nucleotides and amino acids in HA1 was 99.39% and 99.08% 99.04% and 97.78%. In comparison with the A / B risbane / 10/2007 influenza A vaccine recommended by the World Health Organization (WHO) in 2008-2009, the average amino acid homology of the HA1 region of Guangxi isolates was 99.81%, 99.25% and 97.94, respectively %. In 2009 Guangxi A 3 influenza virus vaccine strains compared with the year, HA1 epitopes occurred 6 amino acid substitutions, and occurred in the A, D, E three areas. The distribution of homologous strains and outbreaks in each year were more than 99.00%. In 2007, the average homology of amino acids in HA1 was 99.83% and 99.12%, respectively, compared with those in Shanghai and Beijing. The comparison results in 2008 were 98.93% and 99.25% respectively. Phylogenetic tree analysis showed that the isolates in 2009 were far away from the vaccine strains in 2007, 2008 and current year and branched off separately. Conclusion The characteristics of HA1 gene of influenza A virus in Guangxi in 2007 and 2008 are quite similar. In 2009, there was a great variation in the strain HA1. Guangxi and Shanghai, Zhejiang and other places closer to the isolates, but with a greater difference with the Beijing isolates. The vaccine strains recommended by WHO lagged Guangxi epidemic strains in that year.
