目的对肝纤维化实验动物模型的肝功能和肝纤维化程度等指标进行评估。方法肝纤维化大鼠模型的制备,肝纤维化指标。血清透明质酸(HA),层粘连蛋白(LN),Ⅲ型前胶原(PCⅢ),Ⅳ型胶原(Ⅳ-C)。肝功能指标:①反映肝细胞蛋白合成代谢功能的指标:总蛋白TPROT(g/L),白蛋白ALB(g/L),球蛋白GLB(g/L);②肝细胞受损的指标:谷草转氨酶AST(U/L),谷丙转氨酶ALT(U/L),胆碱脂酶CHE(U/L);③反映肝脏胆排泄、分泌及解毒功能的指标:总胆红素TBILI(μmol/L),直接胆红素DBILI(μmol/L),间接胆红素IBILI(μmol/L),总胆汁酸TBA(μmol/L);④胆汁淤积指示酶:谷氨酰转肽酶GGT(U/L),碱性磷酸酶ALP(U/L)。结果纤维化肝脏的形态学和病理学结果显示的大鼠的肝纤维化处于终末期状态;对于肝纤4项,除层粘连蛋白(LN)外,正常组和肝纤维化组的大鼠有显著性差异;对于肝功15项,除了总蛋白(TPROT)和胆碱酯酶(CHE)外,正常组和肝纤维化组的大鼠有显著性差异。结论肝纤维化大鼠的肝纤维化和肝功指标对对照组相比有显著性差异,与肝纤维化的病理和形态学检测相一致。
Objective To evaluate the liver function and the degree of liver fibrosis in animal models of liver fibrosis. Methods Preparation of liver fibrosis rat model, indicators of liver fibrosis. Serum hyaluronic acid (HA), laminin (LN), type Ⅲ procollagen (PC Ⅲ) and type Ⅳ collagen (Ⅳ-C) were measured. Index of liver function: (1) The indexes reflecting the function of protein synthesis and metabolism of hepatocytes: total protein TPROT (g / L), albumin ALB (g / L) and globulin GLB (U / L), ALT (U / L) and cholinesterase (CHE) in serum of the rats. ③The indexes reflecting the function of excretion of liver, secretion and detoxification: Total bilirubin / L), direct bilirubin DBILI (μmol / L), indirect bilirubin IBILI (μmol / L), total bile acid TBA (μmol / L); ④ cholestasis indicator enzyme: glutamyl transpeptidase GGT U / L), alkaline phosphatase ALP (U / L). Results The morphological and pathological findings of fibrotic liver showed that the hepatic fibrosis of rats was in the terminal state. For the four items of hepatic fibrosis, except the laminin (LN), the rats in normal group and hepatic fibrosis group had There were significant differences between the normal group and the hepatic fibrosis group except for total protein (TPROT) and cholinesterase (CHE) in 15 items of liver function. Conclusion The liver fibrosis and liver function index of liver fibrosis rats have significant difference compared with the control group, which is consistent with the pathological and morphological examination of liver fibrosis.
