目的通过观察Apo-E(-/-)小鼠主动脉根部血管MMP-9的表达情况,进一步了解MMP-9与动脉粥样硬化斑块形成过程的相关性。方法本实验采用免疫组织化学和逆转录聚合酶链技术对MMP-9在基因敲出小鼠动脉中的表达进行研究。结果正常小鼠冠状动脉中MMP-9有微量表达,动脉粥样硬化斑块中检测到MMP-9 mRNA和MMP-9蛋白的表达明显增高。结论 MMP-9可能参与小鼠动脉粥样硬化病变过程,并在小鼠主动脉根部血管壁斑块中表达增高。
Objective To investigate the relationship between the expression of MMP-9 and the formation of atherosclerotic plaque in Apo-E (- / -) mice aorta. Methods In this experiment, the expression of MMP-9 in the artery of gene knockout mice was studied by immunohistochemistry and reverse transcriptase polymerase chain reaction. Results The expression of MMP-9 mRNA and MMP-9 protein in atherosclerotic plaque was significantly increased in normal mice. Conclusion MMP-9 may be involved in the process of atherosclerotic lesions in mice and its expression is increased in plaque in the root of aorta in mice.
