目的观察糖尿病大鼠肾脏组织蛋白激酶C (proteinkinaseC ,PKC)活性变化规律。方法大鼠随机分为正常对照组 (A组 )和糖尿病组 (B组 ) ,分别于 4、6周应用酶联免疫法测定肾脏PKC活性 ;同时测定尿蛋白、肌酐清除率、血肌酐及肾脏肥大指数 ,对肾脏组织进行光镜及电镜观察。结果PKC活性升高与肾质量 /体质量、尿蛋白排泄率呈相关关系 ,且与肾功能下降及组织学改变具有时间一致性。结论PKC活性升高可能是导致糖尿病肾脏病变的机制之一。
Objective To observe the changes of protein kinase C (PKC) activity in kidney of diabetic rats. Methods The rats were randomly divided into normal control group (group A) and diabetic group (group B). The renal PKC activity was measured by enzyme linked immunosorbent assay (ELISA) at 4 and 6 weeks. Urine protein, creatinine clearance rate, serum creatinine and renal Hypertrophy index, the kidney tissue microscopy and electron microscopy. Results The increase of PKC activity correlated with the renal mass / body mass and urinary protein excretion rate, and was consistent with the decline of renal function and histological changes. Conclusions Increased PKC activity may be one of the mechanisms leading to diabetic nephropathy.
