目的 研究内毒素对急性肝损伤大鼠细胞免疫功能的影响及中药丹参与大黄的治疗作用。方法 取Wistar大鼠 30只 ,随机分为三组 :正常组、硫代乙酰胺 (TAA)组和治疗组 (TAA +丹参与大黄组 )。TAA造大鼠急性肝损伤模型。测以下指标 :血浆内毒素 (ET)、肿瘤坏死因子 -α (TNF -α)、白细胞介素 - 2 (IL - 2 )及丙氨酸转氨酶 (ALT)。取肝左叶于 10 %中性福尔马林溶液固定 ,石蜡包埋 ,做组织染色观察。结果 TAA组血浆ET、ALT与TNF -α较正常组明显升高 ,IL - 2较正常组明显下降 ,均有显著性差异 (P <0 0 5 )。治疗组ET、ALT与TNF -α较TAA组下降 ,IL - 2较TAA组升高 ,均有显著性差异 (P <0 0 5 )。血浆ET与TNF -α呈正相关 (γ =0 6 2 8,P <0 0 5 ) ,与IL - 2呈负相关 (γ =- 0 712 ,P <0 0 5 )。结论 急性肝损伤大鼠血浆内毒素水平明显升高 ,同时伴有免疫功能紊乱。丹参与大黄通过降低内毒素水平 ,可调整细胞免疫功能 ,达到治疗的目的。
Objective To study the effect of endotoxin on cellular immune function in rats with acute liver injury and the therapeutic effect of Chinese traditional medicine Salvia miltiorrhiza on rhubarb. Methods Thirty Wistar rats were randomly divided into three groups: normal group, thioacetamide (TAA) group and treatment group (TAA + Danshen and rhubarb group). TAA model of acute liver injury in rats. The following indexes were measured: plasma endotoxin (ET), tumor necrosis factor - α (TNF - α), interleukin - 2 (IL - 2) and alanine aminotransferase (ALT). The left lobe of the liver was fixed in 10% neutral formalin solution and embedded in paraffin for histological observation. Results The levels of plasma ET, ALT and TNF - α in TAA group were significantly higher than those in normal group, while the levels of IL - 2 in normal group were significantly decreased (P <0.05). The levels of ET, ALT and TNF-α in the treatment group were lower than those in the TAA group, while the levels of IL - 2 in the treatment group were significantly higher than those in the TAA group (P <0.05). Plasma ET was positively correlated with TNF-α (γ = 0 6 2 8, P 0 05) and negatively correlated with IL 2 (γ = - 0 712, P 0 05). Conclusion The levels of plasma endotoxin in acute liver injury rats were significantly increased, accompanied by immune dysfunction. Danshen rhubarb by reducing the level of endotoxin, cellular immune function can be adjusted to achieve the purpose of treatment.
