目的探讨结肠癌转移相关基因1(metastasis-associated in colon cancer-1,MACC1)、肝细胞生长因子(HGF)和C-met蛋白在子宫内膜癌中的表达以及其相关的临床病理特点。方法应用免疫组织化学和RT-PCR技术检测2009年1月至2011年7月郑州大学第一附属医院妇产科行手术治疗的子宫内膜癌患者的癌变内膜标本39例及同期25例不典型增生的子宫内膜、25例单纯性增生子宫内膜、25例正常子宫内膜中MACC1、HGF和C-met蛋白、mRNA的表达情况,并分析3者与子宫内膜癌临床病理特征的关系及其相关性。结果 MACC1、HGF和C-met蛋白在子宫内膜癌组织中的阳性率分别为64.1%、61.5%和69.2%,显著高于正常、单纯增生以及不典型增生的内膜组织,差异均有统计学意义(P<0.05)。子宫内膜癌中MACC1、HGF、MET基因的mRNA相对表达量分别为0.216±0.062、0.134±0.054、0.172±0.044,也明显高于其他3组,差异均有统计学意义(P<0.05)。MACC1、HGF和C-met蛋白的异常高表达与子宫内膜癌的临床分期、肌层浸润和淋巴结转移相关,临床分期越晚、肌层浸润越深、伴随淋巴结转移的癌组织中MACC1、HGF和C-met蛋白表达越高(P<0.05)。子宫内膜癌组织中MACC1蛋白的表达与HGF和C-met蛋白的表达呈正相关(r=0.525,P=0.000;r=0.393,P=0.008),HGF蛋白与C-met蛋白表达呈正相关(r=0.334,P=0.027)。结论在子宫内膜癌组织中MACC1具有作为子宫内膜癌诊断、治疗评估分子标志物的潜在价值,MACC1、HGF和C-met蛋白表达异常可能协同参与子宫内膜癌的进展。
Objective To investigate the expression of metastasis-associated in colon cancer-1 (MACC1), hepatocyte growth factor (HGF) and C-met protein in endometrial carcinoma and its clinicopathological features. Methods Immunohistochemistry and RT-PCR were used to detect 39 cases of endometrial carcinoma and 39 cases of endometrial carcinoma underwent surgical treatment in the department of gynecology and obstetrics of the First Affiliated Hospital of Zhengzhou University from January 2009 to July 2011. Typical hyperplasia of endometrium, 25 cases of simple hyperplasia endometrium, 25 cases of normal endometrium MACC1, HGF and C-met protein, mRNA expression, and analysis of three with clinicopathological features of endometrial cancer Relationship and its relationship. Results The positive rates of MACC1, HGF and C-met protein in endometrial carcinoma were 64.1%, 61.5% and 69.2% respectively, which were significantly higher than those in normal, simple hyperplasia and atypical hyperplasia Significance (P <0.05). Abnormally high expression of MACC1, HGF and C-met protein was associated with the clinical stage, myometrial invasion and lymph node metastasis of the endometrial carcinoma. The expression of MACC1 protein in endometrial carcinoma was positively correlated with the expression of HGF and C-met protein (r = 0.525, P = 0.000; r = 0.393, P = 0.008) r = 0.334, P = 0.027). Conclusion MACC1 has potential value as a molecular marker for the diagnosis and treatment of endometrial cancer. The abnormal expression of MACC1, HGF and C-met may be involved in the progress of endometrial cancer.
