目的构建系统的斑马鱼血管内脂代谢的研究方法。方法利用油红O染色、荧光探针标记、直接血脂检测等方法研究斑马鱼胚胎、幼鱼、成鱼等不同发育时期血管内的脂质代谢,以及不同饮食对斑马鱼血脂的影响。结果油红可将斑马鱼胚胎血管内的脂质染色,荧光探针可荧光标记斑马鱼胚胎血管内的胆固醇。喂食蛋黄液可升高斑马鱼幼鱼的血脂水平,油红O染色可反映这种血脂的变化。高胆固醇饮食可升高斑马鱼幼鱼血管内胆固醇水平,荧光探针可标记幼鱼血管内胆固醇的变化。血脂检测显示高脂饮食的斑马鱼血清总胆固醇和甘油三酯均高于普通饮食的斑马鱼。结论本研究利用油红O染色、荧光探针标记及血脂检测等技术构建了斑马鱼不同发育阶段血管内脂代谢的研究方法。
Objective To construct a systematic study of zebrafish vascular lipid metabolism. Methods The lipid metabolism in zebrafish embryos, juveniles and adult fish at different developmental stages was studied by oil red O staining, fluorescent probe labeling and direct blood lipids detection. The effects of different diets on zebrafish blood lipid were also studied. Results Oil red staining of lipid in zebrafish embryo blood vessels, fluorescent probes fluorescence of zebrafish embryo intravascular cholesterol. Feeding egg yolk liquid can increase the lipid level of zebrafish larvae, oil red O staining can reflect the changes of this lipid. High cholesterol diet can increase the level of cholesterol in zebrafish larvae, and fluorescent probes can mark the change of cholesterol in juvenile fish. Lipid test showed that the high-fat diet of zebrafish serum total cholesterol and triglycerides were higher than the normal diet of zebrafish. Conclusion In this study, oil red O staining, fluorescent probe labeling and detection of lipids and other technologies to build zebrafish vascular lipid metabolism in different stages of development methods.
