细胞凋亡紊乱在消化道肿瘤的发生、发展过程中起着重要作用。BNIP3作为Bcl-2蛋白家族中BH3-only亚家族的成员之一,具有BH3结构域和TM跨膜结构区,属于线粒体促凋亡蛋白。BNIP3可通过Caspase非依赖性线粒体凋亡途径诱导细胞死亡,还可以介导自噬性细胞死亡,其表达受缺氧及其他因素的影响。BNIP3在肿瘤中的表达具有组织特异性,在乳腺癌、肺癌和宫颈癌等肿瘤中BNIP3高表达,而在胰腺癌、胃癌和结直肠癌中BNIP3则表达沉默。BNIP3在肿瘤中的表达缺失可导致肿瘤细胞耐受缺氧,对放化疗产生抵抗。全面了解BNIP3在各种肿瘤中的表达谱,可能使BNIP3成为临床指导肿瘤治疗及判断肿瘤预后的新靶标。
Apoptosis disorders in the occurrence and development of gastrointestinal tumors play an important role. As a member of the BH3-only subfamily of the Bcl-2 protein family, BNIP3 has a BH3 domain and a TM transmembrane domain, belonging to mitochondrial proapoptotic proteins. BNIP3 induces cell death via the caspase-independent mitochondrial apoptotic pathway and also mediates autophagic cell death, whose expression is affected by hypoxia and other factors. BNIP3 is tissue-specific in tumors and has a high expression of BNIP3 in tumors such as breast, lung and cervical cancer, while BNIP3 is silenced in pancreatic, gastric and colorectal cancers. The loss of BNIP3 expression in tumors can result in the tumor cells being resistant to hypoxia and resistant to radiotherapy and chemotherapy. A comprehensive understanding of the expression profile of BNIP3 in various tumors may make BNIP3 a new target for clinical treatment of cancer and judgment of tumor prognosis.
