目的:观察健步通络熏蒸液对膝骨性关节炎大鼠血清和关节液中白介素1β(IL-1β)、基质金属蛋白酶3(MMP-3)含量的影响及其相关性,探讨健步通络熏蒸液治疗膝骨性关节炎的作用机制。方法:将32只大鼠随机分为四组,分别为A正常组、B模型组、C对照组、D治疗组,B、C、D三组按照Hulth造模方法造成膝骨性关节炎模型。治疗组给予健步通络熏蒸液治疗,对照组给予扶他林外涂。给药10天后测定大鼠血清和关节液中白介素1β、基质金属蛋白酶3含量。结果:对血清、关节液IL-1β、MMP-3指标比较,治疗组、对照组均低于模型组,差异有统计学意义(P<0·05);模型组明显高于正常对照组,差异有显著的统计学意义(P<0·01);治疗组与对照组在上述指标两组间比较无明显性差异(P>0·05)。结论:采用Hulth造模法可以制造出膝骨性关节炎动物模型;膝骨性关节炎可以导致大鼠血清、关节液中IL-1β、MMP-3含量升高,IL-1β、MMP-3可能是参与0A发病过程的重要介质;健步通络熏蒸液、扶他林均具有降低血清、关节液中IL-1β、MMP-3含量,在一定程度上对膝骨性关节炎有治疗作用。
Objective: To observe the effect of Jianbu Tongluo fumigation fluid on interleukin-1β (IL-1β) and matrix metalloproteinase-3 (MMP-3) in the serum and synovial fluid of knee osteoarthritis rats and to explore its relationship with Jianbu Tongluo Mechanism of treatment of knee osteoarthritis with fumigation fluid. Methods: 32 rats were randomly divided into four groups: A normal group, B model group, C control group, D treatment group, B, C, D three groups according to Hulth model of knee osteoarthritis . The treatment group was given Jianbu Tongluo fumigation treatment, and the control group was given Futailin top coat. After administration for 10 days, the contents of interleukin-1β and matrix metalloproteinase 3 in serum and synovial fluid of rats were determined. Results: Compared with the model group, the levels of IL-1β and MMP-3 in the serum and synovial fluid in the treatment group and the control group were significantly lower than those in the model group (P <0.05); the model group was significantly higher than the normal control group, (P <0.01). There was no significant difference between the two groups in treatment group and control group (P> 0.05). Conclusion: Knee osteoarthritis can be made by Hulth modeling. Knee osteoarthritis can increase the levels of IL-1β, MMP-3 and IL-1β, MMP-3 in serum and synovial fluid May be involved in the pathogenesis of 0A an important medium; Jianbu Tongluo fumigation fluid, Futailin have reduced serum, synovial fluid IL-1β, MMP-3 content, to a certain extent, the treatment of knee osteoarthritis.
