目的通过cellcountingkit-8(CCK-8)检测不同粒径硒化镉(CdSe/ZnS)量子点对人恶性黑素瘤细胞A375、A375.s2以及正常人表皮细胞HaCaT的细胞毒性,研究硒化镉量子点对黑素瘤细胞生长的影响。方法分别取处于对数生长期的A375、A375.s2以及HaCaT细胞铺入96孔板内,次日分别向铺有细胞的孔内加入浓度为162、81、54、40.5、27、10.125、4.05、2.025、1.0125nmol/L水溶性量子点(QDs)-605与QDs-545进行孵育,24h后每孔加入10μlCCK-8,测定2种硒化镉量子点对A375、A375.s2及HaCaT细胞增殖的影响。结果 CCK-8法显示,随着QDs-605与QDs-545浓度的增加,A375和A375.s2细胞存活率随之下降。不同浓度的QDs-605与QDs-545作用于A375、A375.s2细胞,24h后其细胞存活率均低于对照组(P<0.05,P<0.01),但是对HaCaT细胞没有抑制作用。结论硒化镉量子点对A375、A375.s2人黑素瘤细胞有一定的抑制作用,而对HaCaT正常人表皮细胞抑制作用不明显。
Objective To investigate the cytotoxicity of cadmium selenide (CdSe / ZnS) quantum dots on human malignant melanoma cells A375, A375.s2 and normal human epidermal cells HaCaT by cellcounting kit-8 (CCK-8) Effect of Quantum Dots on Melanoma Cell Growth. Methods A375, A375.s2 and HaCaT cells in logarithmic growth phase were plated into 96-well plates. The next day, the cells were plated with cells at concentrations of 162, 81, 54, 40.5, 27, , 2.025,1.0125nmol / L water-soluble quantum dots (QDs) -605 were incubated with QDs-545. After 24h, 10μl of CCK-8 was added into each well, and the proliferation of A375, A375.s2 and HaCaT cells Impact. Results The CCK-8 assay showed that the survival rates of A375 and A375.s2 cells decreased with the increase of QDs-605 and QDs-545 concentrations. The cell viabilities of QDs-605 and QDs-545 cells in A375 and A375.s2 cells treated with different concentrations of QDs-605 and QDs-545 were lower than those in control group (P <0.05, P <0.01), but not in HaCaT cells. Conclusion CdSe QDs can inhibit A375 and A375.s2 human melanoma cells to a certain extent, but not to HaCaT normal human epidermal cells.
