目的 :观察高功率微波 (highpowermicrowave ,HPM)对心肌细胞的影响及细胞内钙离子浓度的变化 ,探讨HPM对心肌细胞损伤的发生机制。方法 :以功率密度 95 0mW /cm2 的HPM辐照心肌细胞后 ,用荧光染料Fluo_3/AM负载 ,于激光扫描共聚焦显微镜下观察心肌细胞形态和 [Ca2 +]i的变化 ;用FITC_膜联蛋白 (annexin)V/PI双染色法 ,于流式细胞仪检测心肌细胞的坏死和凋亡。结果 :HPM辐照后 ,细胞内钙离子浓度明显降低 ,与对照组相比差异显著 (P <0 .0 1)。心肌细胞发生坏死和凋亡 ,与对照组相比差异显著 (P <0 .0 1)。结论 :HPM辐照可引起心肌细胞[Ca2 +]i明显降低 ,提示HPM辐照可引起心肌细胞膜的损伤 ,从而造成 [Ca2 +]i的大量漏出和细胞的死亡。
OBJECTIVE: To observe the effect of high power microwave (HPM) on cardiomyocytes and the change of intracellular calcium concentration, and to explore the mechanism of HPM on cardiomyocyte injury. Methods: Myocardial cells were irradiated with HPM with a power density of 95 0 mW / cm2. Fluorescence dye Fluo_3 / AM was used to load and the changes of myocardial cell morphology and [Ca2 +] i were observed under laser scanning confocal microscopy. Annexin V / PI double staining was used to detect the necrosis and apoptosis of cardiomyocytes in flow cytometry. Results: After irradiation with HPM, the intracellular calcium concentration decreased significantly compared with the control group (P <0.01). Cardiomyocytes necrosis and apoptosis, compared with the control group, significant difference (P <0.01). CONCLUSION: HPM irradiation can cause a marked decrease of [Ca2 +] i in myocardial cells, suggesting that HPM irradiation may cause myocardial cell membrane damage, resulting in massive leakage of [Ca2 +] i and cell death.
