目的:探讨乙酰肝素酶(HPA)反义寡脱氧核苷酸(AS-OND)对人胃癌细胞株SGC-7901细胞基质金属蛋白酶-2(MMP-2)及组织金属蛋白酶抑制剂-2(TIMP-2)表达的影响。方法:采用脂质体介导法将HPAAS-ODN转染SGC-7901细胞G用RT-PCR检测HPA-mRNA表达,用免疫细胞化学的方法检测细胞内MMP-2和TIMP-2的变化。结果:AS-ODN转染48h后GSGC-7901细胞内HPA-mRNA表达下降(P<0.01),MMP-2的表达量亦减少(P<0.01)。结论:HPAAS-ODN可以有效地抑制胃癌细胞MMP-2的表达。
Objective: To investigate the effect of heparanase antisense oligodeoxynucleotide (AS-OND) on the expression of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 TIMP-2) expression. Methods: HPAAS-ODN was transfected into SGC-7901 cells by liposome-mediated method. The expression of HPA-mRNA was detected by RT-PCR and the changes of MMP-2 and TIMP-2 were detected by immunocytochemistry. Results: The expression of HPA-mRNA in GSGC-7901 cells decreased (P <0.01) and the expression of MMP-2 decreased 48h after AS-ODN transfection (P <0.01). Conclusion: HPAAS-ODN can effectively inhibit the expression of MMP-2 in gastric cancer cells.
