本文通过Triton X-100增溶,麦芽糖-淀粉凝胶柱亲和层析等步骤,从猪脂肪细胞表面分离出一类具有凝集素性质的糖蛋白,命名为猪脂肪细胞凝集素(PAL),PAL在凝胶电泳图谱上显示两条蛋白着色带,分子量分别为112,000和89,000,PAL 的血凝活力被麦芽糖、甘露糖、甘露聚糖以及具有类似糖结构的糖蛋白抑制,抑制PAL血凝活力的糖亦阻断肪脂细胞与红血球的粘连.PAL 促使游离脂肪细胞产生集聚,提示位于脂肪细胞表面的PAL可能与脂肪细胞在体内的相互识别和粘连有关。
In this paper, a kind of lectin-like glycoprotein was isolated from the surface of porcine adipose cells by Triton X-100 solubilization, maltose-starch gel column affinity chromatography and so on, named as pig adipocytokinin (PAL) PAL showed two protein bands on the gel electrophoresis map with molecular weights of 112,000 and 89,000, respectively. The hemagglutination activity of PAL was inhibited by maltose, mannose, mannan and glycoproteins with similar glycosidic structures and inhibited PAL blood coagulation activity Of sugar also blocked the adhesion of fat cells to red blood cells.PAL induced the accumulation of free adipocytes, suggesting that the PAL located on the surface of fat cells may be related to the mutual recognition and adhesion of adipocytes in vivo.
