目的··:观察钙拮抗剂盐酸地尔硫对小鼠吗啡戒断反应及血浆和脑匀浆一氧化氮 (NO)含量的影响。方法··:皮下注射定量吗啡 ,建立小鼠吗啡依赖动物模型 ;腹腔注射纳洛酮催促 ,根据小鼠戒断反应中出现跳跃反应的潜伏期、跳跃次数和体重丢失评定戒断反应的强度 ;用硝酸还原法检测其血浆和脑内NO含量。结果·· :3种不同剂量 (25、50、100mg·kg-1)的盐酸地尔硫对小鼠吗啡戒断反应有明显的抑制作用 ,并呈量效关系;100mg·kg-1的盐酸地尔硫可抑制吗啡戒断反应引起的血浆和脑匀浆NO含量的升高。结论·· :盐酸地尔硫可抑制小鼠吗啡戒断反应 ,并能抑制戒断反应引起的血浆和脑匀浆NO含量的升高。
Objective: · To observe the effect of diltiazem hydrochloride, a calcium antagonist, on morphine withdrawal response and nitric oxide (NO) content in plasma and brain homogenates of mice. RESULTS: Diltiazem hydrochloride at 3 different doses (25, 50 and 100 mg · kg -1) significantly inhibited the morphine withdrawal response in mice and showed a dose-response relationship. The effect of 100 mg · kg -1 Diltiazem hydrochloride can inhibit the increase of NO content in plasma and brain homogenate caused by morphine withdrawal response. Conclusion Diltiazem hydrochloride can inhibit morphine withdrawal response in mice and inhibit the increase of NO content in plasma and brain homogenate caused by withdrawal response.
