目的:观察当归补血汤对盐酸苯肼介导的小鼠氧化性溶血的治疗效果,并探讨其对红细胞抗氧化能力的影响。方法:采用盐酸苯肼(60 mg·kg-1)分别于第1,4,7天ip ICR小鼠,诱导氧化性溶血模型。实验设正常对照组、模型组、当归补血汤高、中、低剂量组(10,5,2.5 g·kg-1·d-1)和泼尼松组(0.005 g·kg-1·d-1),每组10只。给药干预后,观察当归补血汤对模型小鼠体重、红细胞数量(RBC)、血红蛋白(Hb)含量、红细胞超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的影响。结果:与正常对照组比较,模型组小鼠外周血红细胞的数量和Hb含量显著下降,MDA增加,SOD活性下降(P<0.01或P<0.05)。相对于模型组,当归补血汤可增加模型小鼠红细胞的数量、提高Hb含量、降低MDA水平以及增强SOD活性(P<0.01或P<0.05)。结论:当归补血汤对盐酸苯肼介导的氧化性溶血具有良好的疗效,其作用机制可能与促进机体造血机能,增强红细胞抗氧化能力和拮抗氧化剂对红细胞的损伤有关。
Objective: To observe the therapeutic effect of Dangguibuxuetang on Phenylhydrazine-mediated oxidative hemolysis in mice and its effect on antioxidant capacity of erythrocytes. Methods: IP ICR mice were treated with Phenylhydrazine Hydrochloride (60 mg · kg -1) on days 1, 4 and 7 to induce oxidative hemolysis. The experimental group was divided into normal control group, model group, Dangguibuxuetang high, medium and low dose groups (10,5,2.5 g · kg -1 · d -1) and prednisone group (0.005 g · kg -1 · d -1) 1), each group of 10. After intervention, the effects of Danggui Buxue Decoction on body weight, RBC, Hb, SOD activity and malondialdehyde (MDA) content of the model mice were observed. Results: Compared with the normal control group, the number of peripheral blood erythrocytes and the content of Hb in the model group decreased significantly, the MDA increased and the SOD activity decreased (P <0.01 or P <0.05). Compared with model group, Dangguibuxuetang can increase the number of red blood cells, increase the content of Hb, decrease the level of MDA and increase the activity of SOD (P <0.01 or P <0.05). CONCLUSION: Dangguibuxuetang has good curative effect on Phenylhydrazine hydrochloride-mediated oxidative hemolysis. Its mechanism may be related to promoting the hematopoietic function, enhancing the anti-oxidation ability of erythrocytes and antagonizing the oxidant’s damage on erythrocytes.
