目的 :观察糖肾通络宁对腹腔注射链脲佐菌素后诱发的糖尿病大鼠早期肾脏病变的影响并探讨其作用机制。方法 :将实验用大鼠随机分为正常对照组、模型对照组、中药高剂量组、中药低剂量组、卡托普利组。分别于实验的第 4、8周取各组的一半大鼠收集标本 ,检测血糖 (PG)、甘油三脂 (TG)、总胆固醇 (TC)、低密度脂蛋白胆固醇 (LDL -CH)、血肌酐 (Scr)、尿白蛋白 (UAE)及肾脏肥大指数 ;免疫组化检测肾内转化生长因子 β1(TGFβ1)、纤维连接蛋白 (FN)和Ⅳ型胶原的表达水平。结果 :第 4、8周时各治疗组PG、TG、TC、LDL -CH、Scr、UAE、肾组织TGFβ1表达、FN、Ⅳ型胶原含量较模型组显著降低。结论 :中药糖肾通络宁对糖尿病肾脏病变有一定保护作用 ,其机制可能是通过降低血糖、血脂 ,降低尿白蛋白排泄率 ,下调DM大鼠肾组织内TGF - β1表达、减少细胞外基质积聚而实现的。
Objective : To observe the effect of Tangshen Tongluoning on early renal lesions in diabetic rats induced by intraperitoneal injection of streptozotocin and explore its mechanism of action. Methods: Rats were randomly divided into normal control group, model control group, traditional Chinese medicine high-dose group, traditional Chinese medicine low-dose group, and captopril group. Blood samples (PG), triglycerides (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-CH), blood were collected at the 4th and 8th weeks of each experiment. Creatinine (Scr), urinary albumin (UAE), and renal hypertrophy index were measured. Expressions of transforming growth factor-β1 (TGFβ1), fibronectin (FN), and type IV collagen were detected by immunohistochemistry. RESULTS: At the 4th and 8th weeks, the PG, TG, TC, LDL-CH, Scr, UAE, TGFβ1 expression, FN, and IV collagen contents in the treatment groups were significantly lower than those in the model group. Conclusion: Traditional Chinese medicine Tang Shen Tong Luo Ning has a certain protective effect on diabetic nephropathy, its mechanism may be through lowering blood glucose, blood lipids, reducing urinary albumin excretion rate, down-regulating TGF-β1 expression in DN rat kidney tissue, reducing extracellular matrix Accumulated and achieved.
