目的研究Hiwi基因mRNA及Hi wi蛋白在肝癌组织中的表达。方法92例肝癌及癌旁组织标本,应用RT-PCR、Western blot及免疫组化方法分别检测肝癌和癌旁组织中Hiwi基因mRNA及蛋白的表达。结果肝癌组织Hiwi基因mRNA和Hiwi蛋白表达量显著高于癌旁组织(P<0.05)。Hiwi蛋白主要在肿瘤和癌旁组织细胞胞浆中呈阳性表达。肝癌组织中Hi wi阳性表达率为65.2%(60/92),显著高于癌旁组织的27.2%(25/92)(P<0.05)。结论Hiwi基因mRNA及蛋白在肝癌组织中均呈高表达,可能在肝癌的发生、发展过程中起重要作用。
Objective To study the expression of Hiwi mRNA and Hi wi protein in hepatocellular carcinoma. Methods 92 cases of hepatocellular carcinoma and adjacent non-cancerous tissue samples were collected. The expression of Hiwi mRNA and protein in hepatocellular carcinoma and adjacent non-cancerous tissues was detected by RT-PCR, Western blot and immunohistochemistry respectively. Results The expression of Hiwi mRNA and Hiwi protein in hepatocellular carcinoma tissues was significantly higher than that in paracancerous tissues (P <0.05). Hiwi protein was mainly expressed in the cytoplasm of tumor and paracancerous tissues. The positive expression rate of Hi wi in HCC tissues was 65.2% (60/92), which was significantly higher than that in adjacent tissues (27.2%, 25/92) (P <0.05). Conclusion Hiwi mRNA and protein are highly expressed in hepatocellular carcinoma, which may play an important role in the occurrence and development of hepatocellular carcinoma.
