目的观察Noggin蛋白对乳腺癌细胞MCF-7增殖、侵袭能力的影响。方法以Noggin重组腺病毒感染乳腺癌细胞株MCF-7,MTT法检测Noggin对MCF-7细胞增殖能力的影响,划痕修复实验及Transwell细胞侵袭实验检测其运动和侵袭能力的改变,Real-time PCR和Western blot检测CXCR4和MMP1基因的表达改变。结果过表达Noggin蛋白的MCF-7细胞增殖能力增强(P<0.05);划痕修复实验提示过表达Noggin蛋白的MCF-7细胞划痕愈合延迟;Transwell细胞侵袭实验提示与空病毒组相比,Noggin组穿膜细胞数显著降低(55±4 vs 25±3,P<0.05)。RT-PCR和Western blot结果:过表达Noggin后,MCF-7细胞中CXCR4和MMP1基因表达下调。结论 Noggin蛋白可抑制乳腺癌细胞MCF-7的运动和迁移,可能与下调CXCR4和MMP1基因的表达相关。
Objective To observe the effect of Noggin protein on the proliferation and invasion of breast cancer cell MCF-7. Methods The effect of Noggin on the proliferation of MCF-7 cells was detected by MTT assay using Noggin recombinant adenovirus. The changes of motility and invasion ability of MCF-7 cells were detected by scratch repair assay and Transwell cell invasion assay. Real-time PCR and Western blot detection of CXCR4 and MMP1 gene expression changes. Results The proliferation of MCF-7 cells over-expressing Noggin protein was enhanced (P <0.05). Scratch repair experiments showed that the wound healing of MCF-7 cells over-expressing Noggin protein was delayed. Transwell cell invasion assay showed that compared with empty virus group, The number of transmembrane cells in the Noggin group was significantly lower (55 ± 4 vs 25 ± 3, P <0.05). The results of RT-PCR and Western blot showed that the expression of CXCR4 and MMP1 was down-regulated in MCF-7 cells after overexpression of Noggin. Conclusion Noggin can inhibit the movement and migration of MCF-7 in breast cancer cells, which may be related to the down-regulation of CXCR4 and MMP1 gene expression.
