采用钙荧光探针Fura-2/AM观察了颅通定对培养的猪肺动脉平滑肌细胞胞浆游离钙浓度([Ca2+]i)的影响。结果发现:颅通定可显著抑制高钾和BayK8644所致的[Ca+]i增加,并且有剂量一效应关系,对静息状态[Ca2+]i无明显影响,其作用与维拉帕米相似,但较弱。提示:颅通定降低平滑肌细胞[Ca2+]i的作用与其抑制电压依赖性钙通道有关,此为其降压作用机制之一。
Fura-2 / AM, a calcium fluorescent probe, was used to observe the effects of craniotome on cytosolic free calcium concentration ([Ca2 +] i) in cultured porcine pulmonary artery smooth muscle cells. The results showed that craniotonging could significantly inhibit the increase of [Ca +] i induced by high potassium and BayK8644, and had a dose-effect relationship, which had no significant effect on resting [Ca2 +] i and its effect was similar to that of verapamil, But weaker. It is suggested that cranialtum surely reduces the [Ca2 +] i of smooth muscle cells, which is related to its inhibition of voltage-dependent calcium channel, which is one of the mechanisms of its antihypertensive effect.
