目的:了解2008年5~9月广州市卫生行业从业人员中德尔卑沙门菌的耐药情况,并对菌株进行脉冲场凝胶电泳(PFGE)基因分型,探索广州市从业人员中德尔卑沙门菌的分子分型多态性。方法:从健康体检的人群中分离并鉴定为德尔卑沙门菌,对鉴定出的13株菌株,选择18种抗生素用纸片法进行药物敏感试验;裂解菌细胞并释放全基因组DNA,经限制性内切酶Xba I酶切,用脉冲场凝胶电泳进行分子分型,使用Quantity-One TM软件成像并使用B ioNum erics4.0软件对其同源性进行分析比较。结果:13株沙门菌对9种抗生素敏感,对其余抗生素出现中敏或耐药。对四环素耐药率最高(76.9%),其次为氯霉素(30.8%),复方新诺明(15.4%),氧氟沙星和氨苄西林(7.7%)。13株受试菌株的DNA分子分型图谱,经软件分析,有两株相似性为100%,其他菌株相似性在73.7%和93.8%之间。结论:德尔卑沙门菌对头孢类抗生素完全敏感,对其他抗生素存在中敏或耐药;菌株之间有较高的同源性。
OBJECTIVE: To understand the drug resistance of Salmonella delphi in Guangzhou health industry from May to September in 2008, and to genotype the strains by pulsed-field gel electrophoresis (PFGE) Molecular typing of bacteria Polymorphism. Methods: Salmonella delphi was isolated and identified from 13 healthy volunteers. Thirteen strains were identified and 18 antibiotics were selected for drug susceptibility testing by paper method. The bacterial cells were lysed and whole genome DNA was released. The endonuclease Xba I was digested and subjected to molecular typing using pulsed-field gel electrophoresis and imaged using Quantity-One ™ software and analyzed for homology using the BioNum erics 4.0 software. Results: Thirteen Salmonella strains were sensitive to nine antibiotics and the other antibiotics were sensitive or resistant. The highest rate of resistance to tetracycline (76.9%) was followed by chloramphenicol (30.8%), cotrimoxazole (15.4%), ofloxacin and ampicillin (7.7%). The DNA molecular typing patterns of 13 tested strains were 100% for the two strains and 73.7% and 93.8% for other strains, respectively. Conclusion: Salmonella delphi is completely sensitive to cephalosporins and is sensitive or resistant to other antibiotics. There is a high homology among strains.
