头孢唑啉钠为抗生素类药,用于溶血性链球菌,草绿色链球菌,葡萄球菌、肺炎球菌、淋球菌、大肠杆菌,奇异变形杆菌、痢疾杆菌等感染。目前国内对该药品含量测定方法,均用微生物杯碟法进行测定,考虑微生物杯碟法,繁杂,要制备培养基,缓冲液,需要115℃高压灭菌30分钟,还要制备鉴定用菌液,有的菌液至少要24小时,所用一切器皿也需在160℃干热灭菌二小时,整个试验麻烦、费时。本文参考文献确立了用高效液相色谱法,对头孢唑啉钠直接测定方法,以pH 3.6磷酸盐缓冲液和乙腈混合液为流动相,水扬酸为内标,以十八烷基硅烷键合硅胶为层析柱,紫外检测。方法简便、快速、准确。对九批试样测定结果与微生物杯碟法结果比较无显著性差异。
