目的通过观察运动对慢性应激大鼠不同脑区5-羟色胺(5-HT)含量及外周血Th1/Th2型细胞因子的影响,探讨运动减缓慢性应激损伤的神经免疫学机制。方法 40只雄性SD大鼠随机分为对照组、模型组、运动+模型组及运动组。采用慢性束缚方式建立心理应激模型;采用无负重游泳作为体力运动方式。用高效液相色谱法检测海马、枕叶皮层5-HT含量,并以酶联免疫吸附(ELISA)法检测血清干扰素-γ(IFN-γ)和白细胞介素-4(IL-4)水平以及IFN-γ/IL-4比值变化。结果与对照组比较,模型组海马、枕叶皮层区5-HT含量明显降低(P﹤0.01),血清IFN-γ及IFN-γ/IL-4比值显著下降(P﹤0.01)。与模型组比较,运动+模型组不同脑区5-HT含量明显增加,IFN-γ水平和IFN-γ/IL-4比值明显增高(P﹤0.05)。与对照组比较,运动组不同脑区5-HT升高,差异有统计学意义(P﹤0.05)。结论体力运动可通过增强中枢5-HT系统功能及维持机体的免疫稳态,降低慢性应激对机体神经免疫功能的损害。
Objective To observe the effects of exercise on the levels of 5-HT in peripheral blood and the levels of Th1 / Th2 cytokines in peripheral blood of chronic stress rats and to explore the neuroimmunological mechanism of exercise-induced chronic stress injury. Methods Forty male SD rats were randomly divided into control group, model group, exercise + model group and exercise group. Chronic restraint was used to establish psychological stress model; swimming without load was adopted as physical exercise mode. The contents of 5-HT in the hippocampus and occipital cortex were detected by high performance liquid chromatography (HPLC), and the levels of serum IFN-γ and IL-4 were detected by enzyme linked immunosorbent assay (ELISA) And IFN-γ / IL-4 ratio changes. Results Compared with the control group, the content of 5-HT in the hippocampus and occipital lobe of the model group was significantly decreased (P <0.01) and the ratio of serum IFN-γ and IFN-γ / IL-4 was significantly decreased (P <0.01). Compared with the model group, the content of 5-HT in the brain regions of exercise + model group increased significantly, and the IFN-γ level and IFN-γ / IL-4 ratio increased significantly (P <0.05). Compared with the control group, exercise group 5-HT in different brain regions increased, the difference was statistically significant (P <0.05). Conclusion Physical exercise can reduce the damage of neurological immune function by chronic stress by enhancing the function of central 5-HT system and maintaining the immune homeostasis.
