[目的]探讨微小RNA(miR)-125b对肺癌细胞95D生物学功能的影响。[方法]以95D细胞为实验对象,分别转染miR-125b模拟物和模拟物阴性对照,未经特殊处理为空白组。应用MTT法、流式细胞仪和Transwell小室法分别检测miR-125b表达改变对95D细胞增殖、凋亡、周期及侵袭的影响。[结果]95D细胞转染48 h后,转染组miR-125b的水平提高,表达量为阴性对照组的22.32倍。与阴性对照组相比,miR125b模拟物转染组细胞凋亡率明显降低[(13.77±-0.52)%,(9.90±1.33)%,P<0.05],细胞增殖、周期、侵袭无显著改变(P>0.05)。[结论]miR-125b基因过表达能够抑制肺癌95D细胞凋亡。
[Objective] To investigate the effect of microRNA (miR) -125b on the biological function of lung cancer cell line 95D. [Method] 95D cells were used as experimental subjects and transfected with miR-125b mimic and mock negative control, respectively, without any special treatment. The effects of miR-125b expression on proliferation, apoptosis, cycle and invasion of 95D cells were detected by MTT assay, flow cytometry and Transwell chamber assay. [Results] The expression level of miR-125b in transfected group increased after 48 hours of transfection with 95D cells, which was 22.32 times of that in the negative control group. Compared with the negative control group, the apoptosis rate of miR125b mock transfected group was significantly lower than that of the negative control group [(13.77 ± -0.52)%, (9.90 ± 1.33)%, P <0.05] P> 0.05). [Conclusion] Overexpression of miR-125b can inhibit 95D cell apoptosis in lung cancer.
