目的:评价经腺病毒介导的血管内皮细胞生长因子(VEGF)基因转染后的大鼠骨髓基质干细胞(BMSCs)诱导成内皮细胞的可行性。方法:从大鼠骨髓中分离培养获取BMSCs,分别经Adv-VEGF、Adv-GFP转染,Western印迹分析检测VEGF蛋白的表达情况,细胞生长曲线测定BMSCs增殖特性,同时进行免疫表型鉴定,并与未转染细胞组比较。结果:AdvVEGF转染组BMSCs可分泌VEGF蛋白,而其余两组不分泌VEGF蛋白。转染VEGF基因后,BMSCs生长速度变快,向内皮细胞分化。与Adv-VEGF转染组BMSCs相比,Adv-GFP转染细胞组、未转染细胞组的BMSCs生长速度较慢,未有向内皮细胞分化的证掘。结论:腺病毒介导的VEGF基因转染可诱导BMSCs分化成内皮细胞。
OBJECTIVE: To evaluate the feasibility of adenovirus-mediated induction of endothelial cell growth factor (VEGF) into rat bone marrow stromal stem cells (BMSCs) into endothelial cells. Methods: BMSCs were isolated and cultured from bone marrow of rats. The expression of VEGF protein was detected by Adv-VEGF, Adv-GFP transfection and Western blot analysis. The proliferation characteristics of BMSCs were measured by cell growth curve, and immunophenotype was identified Compared with untransfected cells. Results: BMSCs in AdvVEGF transfection group secreted VEGF protein, while the other two groups did not secrete VEGF protein. Transfection of VEGF gene, BMSCs grow faster, to endothelial cells. Compared with BMSCs transfected with Adv-VEGF, BMSCs in Adv-GFP transfected and untransfected cells grew slowly and did not differentiate into endothelial cells. Conclusion: Adenovirus-mediated VEGF gene transfection can induce BMSCs to differentiate into endothelial cells.
