目的观察火绒草水提取物对2型糖尿病(T2DM)大鼠海马区神经元tau蛋白过度磷酸化水平和学习记忆的影响。方法采用高糖高脂饮食诱导联合链脲佐菌素(STZ)腹腔注射建立T2DM大鼠模型。将健康Wistar雄性大鼠30只,随机均分为对照组、T2DM组和火绒草组。水迷宫实验检测大鼠学习记忆功能;免疫组化法检测海马CA1,CA3,DG区神经元磷酸化tau蛋白p-tau(ser396)表达水平。结果与对照组比较,T2DM组水迷宫逃避潜伏期延长(P<0.01),跨过原平台位置次数减少(P<0.01);免疫组织化学检测海马CA1,CA3,DG区神经元胞浆内ser396表达水平显著上调。与T2DM组比较,火绒草组水迷宫逃避潜伏期缩短(P<0.05),跨过原平台位置次数增多(P<0.05);海马区神经元ser396表达水平下降。结论火绒草能减少海马区神经元过度磷酸化tau蛋白的表达,改善学习记忆功能。
Objective To observe the effects of bletilla striatum aqueous extract on tau hyperphosphorylation and learning and memory in hippocampal neurons of type 2 diabetic rats. Methods T2DM rats were induced by intraperitoneal injection of streptozotocin (STZ) induced by high glucose and high fat diet. Thirty healthy Wistar male rats were randomly divided into control group, T2DM group and Leonurus group. The expression of phosphorylated tau protein p-tau (ser396) in hippocampal CA1, CA3 and DG neurons was detected by water maze test. Results Compared with the control group, the escape latency of water maze in T2DM group was prolonged (P <0.01), and the number of escape across the original platform decreased (P <0.01). The expression of ser396 in cytoplasm of hippocampal CA1, CA3 and DG neurons was detected by immunohistochemistry Significantly increased levels. Compared with T2DM group, the evapotranspiration latency of Tenebaceae group decreased (P <0.05) and the number of crosses over the original platform increased (P <0.05). Ser396 expression in hippocampal neurons decreased. Conclusion Fire velvet can reduce the expression of tau hyperphosphorylated neurons in hippocampus and improve learning and memory.
