目的:对HPLC-DAD-MS对黄连生物碱成分进行分析的效果进行评价分析,为今后的研究工作提供有价值的参考信息。方法:色谱柱选择Zorbax SB C18色谱柱,流动相A选择乙腈,流动相B选择含20mmol/l乙酸铵与浓度0.5%乙酸的水溶液,进行梯度洗脱,检测波长选择345nm。与电喷雾-离子阱质谱检测器联合,对主要色谱峰成分进行鉴定。结果:实验中建立了对黄连中黄连碱、小檗碱、巴马汀三个成分含量进行测定的高效液相色谱法,结果得出,黄连碱、小檗碱、巴马汀的线性范围分别为0.041-0.083ug、0.049±1.01ug、0.045-0.902ug。经LC-MS测定,发现黄连中7个色谱峰。结论:经上述建立的色谱方法,可以对黄连生物碱成分进行检测,方法简便、结果准确可靠,值得推广。
OBJECTIVE: To evaluate and analyze the effect of HPLC-DAD-MS on the analysis of the constituents of Coptis alkaloids and provide valuable reference information for future research. Methods: Zorbax SB C18 column was used as the mobile phase. Acetonitrile was selected as the mobile phase A and mobile phase B was selected as the mobile phase B. The mobile phase B consisted of 20 mmol / l ammonium acetate and 0.5% acetic acid in water. In combination with electrospray ion trap mass spectrometry, the main peak components were identified. Results: The HPLC method was established for the determination of berberine, berberine and palmatine in the experiment. The results showed that the linear ranges of berberine, berberine and palmatine were 0.041-0.083ug, 0.049 ± 1.01ug, 0.045-0.902ug. After LC-MS determination, found seven peaks in Coptis. Conclusion: The chromatographic method established above can detect the constituents of Coptis chinensis alkaloids, the method is simple, the results are accurate and reliable, and it is worth to be popularized.
