目的为探讨麻疯树的性别分化情况,筛选出麻疯树雌花和花芽的差异表达基因。方法以麻疯树雌花为驱动子,花芽为检测子,使用SSH法进行正向抑制消减杂交。结果得到完整性较好的总RNA,扩增出的双链cDNA效果良好;通过平板筛选,获得2 000余个单克隆;结果表明,用SSH法能有效构建麻疯树花的差异表达基因的差减文库。结论:成功构建了麻疯树花的差减文库。
Objective To investigate the gender differentiation of Jatropha curcas and screen the differentially expressed genes of Jatropha curcas and flower buds. Methods The female flower of Jatropha curcas was used as the driving force, the flower bud was used as the detector, and SSH was used to inhibit the hybridization. The results showed that the complete RNA was better and the double-stranded cDNA was amplified well. More than 2,000 single clones were obtained by screen screening. The results showed that SSH could effectively construct differentially expressed genes of Jatropha curcas Subtraction library. Conclusion: A subtractive library of Jatropha curcas was constructed successfully.
