目的探讨肝细胞生长因子(HGF)在白血病儿童血清及骨髓间充质干细胞(MSCs)培养上清液中的表达及意义。方法采用细胞贴壁法获取足量的MSCs,通过荧光倒置显微镜观察细胞形态及流式细胞仪检测免疫表型的方法对MSCs进行鉴定,应用酶联免疫吸附实验检测白血病患儿和非感染血液病儿童(对照组)的外周血清及白血病患儿MSCs培养上清液中的HGF水平。结果白血病初诊及复发患儿MSCs培养上清液中HGF的浓度为849.98(564.04~3 113.7)pg/m L,而白血病完全缓解期患儿为290.00(83.54~644.27)pg/m L,差异有统计学意义(P<0.05)。白血病初诊及复发患儿血清中HGF浓度为1 172.03(464.24~5 134.40)pg/m L,白血病完全缓解期患儿血清中HGF浓度为324.38(98.25~785.76)pg/m L,对照组血清中HGF浓度为301.77(78.25~607.44)pg/m L,白血病初诊及复发患儿血清中HGF浓度高于白血病完全缓解期患儿及对照组,(P<0.05);而白血病完全缓解期患儿与对照组血清中的HGF浓度相比差异无统计学意义(P>0.05)。结论白血病患儿外周血清中能检测到HGF的异常表达,HGF水平与白血病病情、疗效有一定的相关性。体外培养的白血病患儿骨髓MSCs能分泌HGF,其中来自白血病初诊患儿的MSCs分泌HGF尤为显著,提示HGF的高分泌参与白血病骨髓微环境的功能异常。
Objective To investigate the expression and significance of hepatocyte growth factor (HGF) in the supernatant of leukemia serum and bone marrow mesenchymal stem cells (MSCs). Methods MSCs were obtained by cell attachment method. MSCs were identified by fluorescence inverted microscope and immunophenotyping by flow cytometry. Enzyme-linked immunosorbent assay was used to detect leukemia and non-infected blood diseases Children (control group) of peripheral blood and leukemia MSCs culture supernatant HGF levels. Results The concentration of HGF in MSCs culture supernatant of newly diagnosed and relapsed children with leukemia was 849.98 (564.04 ~ 3 113.7) pg / m L, while it was 290.00 (83.54 ~ 644.27) pg / m L in children with complete remission of leukemia Statistical significance (P <0.05). The concentration of HGF in serum of newly diagnosed and relapsed children with leukemia was 1 172.03 (464.24 ~ 5 134.40) pg / m L, and the concentration of HGF in children with complete remission of leukemia was 324.38 (98.25 ~ 785.76) pg / m L, HGF concentration was 301.77 (78.25 ~ 607.44) pg / m L, serum HGF concentration in newly diagnosed and relapsed children with leukemia was higher than that in children with complete remission of leukemia and control group (P <0.05); while in children with complete remission of leukemia There was no significant difference in HGF concentration in the control group (P> 0.05). Conclusion Abnormal expression of HGF can be detected in the peripheral blood of children with leukemia, and the level of HGF has some correlation with the disease and curative effect of leukemia. In vitro cultured MSCs from leukemia children can secrete HGF, among which MSCs from newly diagnosed leukemia patients are especially significant, suggesting that HGF secretion is involved in the dysfunction of leukemia bone marrow microenvironment.
