目的探讨外源性转化生长因子β1(TGF-β1)抗体对SiO2刺激的小鼠肺成纤维细胞生长的抑制作用及细胞周期的影响。方法实验分为3组:空白对照,SiO2(100 mg/L)组,SiO2(100 mg/L)+TGF-β1(1.0μg/ml)抗体组。应用MTT法检测小鼠肺成纤维细胞增殖的变化,流式细胞仪检测细胞周期。结果各处理组细胞增殖差异有统计学意义(P<0.05),SiO2组细胞增殖最明显,其次为SiO2+TGF-β1抗体组;与空白对照组比较,SiO2组G0/G1期细胞百分率显著降低,S期及G2/M期细胞百分率增加,差异均有统计学意义(P<0.05),经TGF-β1抗体处理后,G0/G1期细胞百分率与SiO2组比较明显增加,S、G2/M期细胞百分率显著降低(P<0.05)。结论 TGF-β1抗体可能通过阻止SiO2刺激的小鼠肺成纤维细胞由G0/G1期进入S期,从而抑制肺成纤维细胞增殖。
Objective To investigate the inhibitory effect of exogenous transforming growth factor β1 (TGF-β1) antibody on the growth of mouse lung fibroblasts stimulated with SiO2 and the cell cycle. Methods The experiment was divided into three groups: blank control, SiO2 (100 mg / L), SiO2 (100 mg / L) and TGF-β1 (1.0μg / ml) antibody group. The proliferation of mouse lung fibroblasts was detected by MTT assay and the cell cycle was detected by flow cytometry. Results The proliferation of cells in each treatment group was significantly different (P <0.05). The proliferation of cells in SiO2 group was the most obvious, followed by SiO2 + TGF-β1 antibody group. Compared with the blank control group, the percentage of cells in G0 / G1 phase was significantly decreased (P <0.05). After treatment with TGF-β1 antibody, the percentage of cells in G0 / G1 phase increased significantly compared with that in SiO2 group, but the percentage of cells in S, G2 / M phase increased significantly The percentage of senescent cells was significantly decreased (P <0.05). Conclusion TGF-β1 antibody may inhibit the proliferation of lung fibroblasts by inhibiting the progression of SiO2-stimulated mouse lung fibroblast from G0 / G1 phase to S phase.
