目的:研究X染色体连锁凋亡抑制基因(XIAP)小分子抑制剂Embelin在体外对宫颈癌细胞株He La生长的影响,并探讨其作用机制。方法:应用CCK-8检测不同浓度Embelin对宫颈癌细胞株He La增殖的影响;利用流式细胞仪检测Embelin对He La细胞凋亡的影响;Western blot方法检测Embelin作用后细胞XIAP、Caspase-3、Caspase-9表达的变化。结果:Embelin对宫颈癌细胞株He La增殖具有显著抑制作用,差异有统计学意义(P<0.05);经不同浓度Embelin处理后,He La凋亡率明显增高,与对照组比较差异有统计学意义(P<0.01);经Embelin作用24 h后,He La细胞Caspase-9,Caspase-3,PARP被激活,表达增加。结论:Embelin在体外可显著抑制He La细胞增殖,诱导细胞凋亡,其机制可能是通过拮抗XIAP的作用,激活Caspase依赖性的细胞凋亡途径。
AIM: To investigate the effect of Embellin, a small molecule inhibitor of X-linked inhibitor of apoptosis (XIAP) on the growth of cervical cancer cell line He La in vitro and to explore its mechanism. Methods: The effect of different concentrations of Embelin on the proliferation of cervical cancer cell line He La was examined by CCK-8. The effect of Embelin on the apoptosis of He La cell was detected by flow cytometry. The expression of XIAP, Caspase-3 , Caspase-9 expression changes. Results: Embelin had a significant inhibitory effect on the proliferation of cervical cancer cell line He La, the difference was statistically significant (P <0.05). After treated with different concentrations of Embelin, the apoptosis rate of He La was significantly higher than that of the control group (P <0.01). Caspase-9, Caspase-3 and PARP were activated in HeLa cells after Embelin treatment for 24 h. CONCLUSION: Embelin can significantly inhibit Hela cell proliferation and induce apoptosis in vitro. The mechanism may be that Caspase-dependent apoptosis pathway may be activated by antagonizing the effect of XIAP.
