目的研究大鼠皮肤肌肉切口牵拉术(SMIR)对切口周围组织神经生长因子(NGF)mRNA表达的影响。方法 36只雄性SD大鼠随机均分为假手术组、SMIR组和空白对照组。假手术组只切开皮肤、肌肉;SMIR组切开皮肤、肌肉后牵拉1h制作SMIR模型;空白对照组不做任何处理。术前和术后第1、3、5、10、14、21、28天分别测量各组机械缩足阈值(MWT),术后第3天RT-PCR检测各组切口周围肌肉组织NGF mRNA的表达。结果 SMIR组术后第1-28天MWT较空白对照组下降(P<0.05),术后第3-28天较假手术组下降(P<0.05)。SMIR组术后第3天切口周围肌肉组织NGF mRNA表达较空白对照组和假手术组增加(P<0.01),假手术组NGF mRNA表达较空白对照组增加(P<0.05)。结论 SMIR促进术后机械性痛觉过敏和NGF mRNA表达上调;NGF可能参与SMIR术后疼痛的发生、发展。
Objective To study the effect of SMIR on the expression of nerve growth factor (NGF) mRNA in the tissue around the incision. Methods Thirty-six male Sprague-Dawley rats were randomly divided into sham operation group, SMIR group and blank control group. Sham operation group only cut skin and muscle; SMIR group cut skin, muscle stretch 1h after making SMIR model; blank control group without any treatment. The mechanical contracting threshold (MWT) of each group was measured preoperatively and on the 1st, 3rd, 5th, 10th, 14th, 21st, 28th day after operation. The mRNA expression of NGF in the muscle tissue around the incision expression. Results The MWT of SMIR group decreased from the first day to the 28th day after operation (P <0.05), and decreased from the 3rd to 28th day after operation (P <0.05). The expression of NGF mRNA in the muscle tissue around the incision in the SMIR group was higher than that in the blank control group and the sham operation group on the 3rd day after operation (P <0.01). The expression of NGF mRNA in the sham operation group was higher than that in the blank control group (P <0.05). Conclusion SMIR can promote mechanical hyperalgesia and up-regulation of NGF mRNA expression. NGF may be involved in the occurrence and development of postoperative pain in SMIR.
