目的:探讨表面分子在自然杀伤(NK)细胞活化中的信号分子作用及其机制。方法:采用多种粘附分子单抗及配体刺激NK细胞,ELISA法定量检测刺激后NK细胞释放的γ干扰素(IFN-γ)作为NK细胞活化指标,观察各种粘附分子在NK细胞释放IFN-γ中的信号作用。结果:单一CD45单抗刺激即可使NK细胞活化;两种分别为IgG1,IgG2a亚类的CD45单抗及IgG2a亚类CD45单抗的F(ab)2均体现对NK细胞的独立刺激作用。结论:CD45分子介导的NK细胞活化与CD16-Fc段间相互作用无关,CD45分子交联是NK细胞活化的独立刺激信号
OBJECTIVE: To investigate the role of surface molecules in the activation of natural killer (NK) cells and its mechanism. Methods: NK cells were stimulated by a variety of adherent monoclonal antibodies and ligands, and the IFN-γ released by NK cells was detected quantitatively by ELISA as the NK cell activation index. The effects of various adhesion molecules on NK cells Release of IFN-γ in signaling. Results: NK cells were activated by stimulation with a single anti-CD45 monoclonal antibody. F (ab) 2, two anti-CD45 monoclonal antibodies of IgG1 and IgG2a subclasses and CD45 monoclonal antibodies of IgG2a subtype, respectively, showed independent stimulation of NK cells . CONCLUSION: The activation of NK cells mediated by CD45 molecules has nothing to do with the interaction between CD16-Fc segments. The CD45 molecule cross-linking is an independent stimulatory signal of NK cell activation
