以MTS染色法测定实验剂量的Lipofectin对细胞的毒性作用。以 β 半乳糖苷酶基因为报告基因 ,通过Lipofectin而转染 ,用X gal染色法 ,测定转染效率。结果表明实验剂量 (10 μg/ml)的Lipofectin对细胞生长无明显毒性。Lipofectin对多数肾细胞癌细胞的转染是有效的 ,且转染效率随Lipofectin浓度的增高 (2 .5~ 10 μg/ml)而增高。说明Lipofectin可安全、简便地提高肾细胞癌细胞DNA转染效率
The cytotoxic effect of experimental dose of Lipofectin on cells was measured by MTS staining. The β-galactosidase gene was used as a reporter gene and transfected with Lipofectin. The transfection efficiency was measured by X gal staining. The results showed that experimental dose (10 μg/ml) of Lipofectin had no obvious toxicity to cell growth. Lipofectin is effective for the transfection of most renal cell carcinoma cells, and the transfection efficiency increases with the increase of Lipofectin concentration (2.5 to 10 μg/ml). Explains that Lipofectin can safely and simply increase the efficiency of DNA transfection of renal cell carcinoma cells
