目的研究1,2,3-三磷酸肌醇[Ins(1,2,3)P3]对四氧嘧啶所致糖尿病小鼠的降血糖作用。方法用小麦麸皮植酸酶部分水解植酸,水解产物经717型阴离子交换树脂吸附,0.05~0.7mol.L-1盐酸梯度洗脱分离得到Ins(1,2,3)P3。将四氧嘧啶导致的糖尿病小鼠随机分成5组,每天分别ip生理盐水、10、50、100mg.kg-1.d-1的Ins(1,2,3)P3钠盐、盐酸苯乙双胍70mg.kg-1.d-1。连续注射12d,末次空腹眼眶取血测血糖。结果Ins(1,2,3)P3可降低四氧嘧啶所致糖尿病小鼠的血糖值,在浓度为50mg.kg-1.d-1与100mg.kg-1.d-1剂量时,显著降低糖尿病小鼠血糖水平,对正常小鼠血糖无影响。Ins(1,2,3)P3可提高四氧嘧啶型糖尿病小鼠及正常小鼠的胸腺与脾脏指数,抵抗四氧嘧啶引起的小鼠胸腺、胰腺萎缩。结论Ins(1,2,3)P3对糖尿病小鼠具有降血糖作用。
Objective To study the hypoglycemic effect of 1,2,3-triphosphate (Ins (1,2,3) P3] on alloxan-induced diabetic mice. Methods Partial hydrolysis of phytic acid with wheat bran phytase was carried out. The hydrolyzate was adsorbed on 717 anion exchange resin and eluted with a gradient of 0.05-0.7 mol·L -1 hydrochloric acid to obtain Ins (1,2,3) P3. Alloxan-induced diabetic mice were randomly divided into 5 groups, ip saline, 10,50,100 mg.kg-1.d-1 Ins (1,2,3) P3 sodium salt daily, phenformin hydrochloride 70 mg.kg-1.d-1. Continuous injection of 12d, the last fasting orbital blood test for blood sugar. Results Ins (1, 2, 3) P3 reduced the blood glucose levels of alloxan-induced diabetic mice significantly at doses of 50 mg.kg-1.d-1 and 100 mg.kg-1.d-1 Reduce blood glucose levels in diabetic mice, normal mice had no effect on blood glucose. Ins (1,2,3) P3 can increase the thymus and spleen index of alloxan-induced diabetic mice and normal mice and resist alloxan induced thymus and pancreatic atrophy in mice. Conclusion Ins (1,2,3) P3 has hypoglycemic effect on diabetic mice.
