目的优化并建立螺旋藻片中β-胡萝卜素的高效液相色谱快速测定方法。方法样品以甲醇∶丙酮(V/V)=1∶3超声提取,用Symmetry Column C18(4.6×150mm,5μm)分析,流动相为纯甲醇,检测波长450nm,流速1.2mL/min。结果该法在10~100mg/L范围内线性关系良好,r=0.9998。平均加标回收率93.7%~101.4%,RSD为0.93%~1.34%(n=6),方法检出限为0.5mg/kg(S/N=3)。结论本法简单、准确、灵敏度高,适合螺旋藻片中β-胡萝卜素的含量的快速测定。
Objective To optimize and establish a rapid method for the determination of β-carotene in Spirulina tablets by high performance liquid chromatography. Methods The samples were extracted with methanol / acetone (V / V) = 1: 3 and analyzed by Symmetry Column C18 (4.6 × 150 mm, 5 μm). The mobile phase was pure methanol. The detection wavelength was 450 nm and the flow rate was 1.2 mL / min. Results The method in the 10 ~ 100mg / L range of good linear relationship, r = 0.9998. The average recovery was 93.7% ~ 101.4% with RSD of 0.93% ~ 1.34% (n = 6). The detection limit was 0.5 mg / kg (S / N = 3). Conclusion This method is simple, accurate and sensitive. It is suitable for the rapid determination of β-carotene in spirulina tablets.
