目的查明引起食物中毒的致病菌,分析菌株间的亲缘关系,为明确食物中毒诊断提供依据。方法采用定量PCR快速筛检、GB法分离、鉴定致病菌;用PFGE对病原菌进行同源性分析。结果从16份蛋糕标本中检出8株肠炎沙门菌,32份患者粪便标本中检出17株肠炎沙门菌,PCR核酸阳性与GB法分离到菌株完全一致;PFGE条带显示患者与蛋糕中检出的肠炎沙门菌带型一致,具有高度同源性。结论本起食物中毒由肠炎沙门菌污染蛋糕所致;PCR法与GB法联合检测有助于快速锁定食物中毒致病菌;PFGE检测发现蛋糕与患者检出的肠炎沙门菌其来源为同一克隆,从基因上证明了食物病原的相关性。
Objective To identify the pathogenic bacteria that cause food poisoning, analyze the genetic relationship among the strains and provide evidence for the diagnosis of food poisoning. Methods Rapid screening by quantitative PCR and GB method were used to isolate and identify pathogenic bacteria. The homology analysis of pathogenic bacteria by PFGE was carried out. Results Eight strains of Salmonella enteritidis were detected in 16 samples of cake and 17 strains of Salmonella enteritidis were detected in stool samples of 32 patients. PCR nucleic acid was completely identical to that isolated by GB method. The PFGE band showed that the patients and the cake were seized Salmonella enteritidis consistent with the type, with a high degree of homology. Conclusion The food poisoning caused by Salmonella enteritidis contamination of the cake caused by PCR; PCR method combined with the GB method will help to quickly lock the food poisoning pathogens; PFGE test found that the cake and the patient’s Salmonella enteritidis from the same clone, Genetically proved the relevance of food pathogens.
