目的研究小鼠孕期暴露于邻苯二甲酸酯与胚胎发育的相关性。方法将60只雌性小鼠随机分为6组,两两配对为对照组1和实验组1、对照组2和实验组2、对照组3和实验组3,其中对照组于假孕第一天即经口给予玉米油,实验组则给予500 mg/kg/d的邻苯二甲酸二乙基己酯(DEHP)和邻苯二甲酸二丁酯(DBP)混合液,通过体外受精的方式获得优质囊胚,移植到假孕小鼠子宫,分别于孕5.5天和9.5天,统计植入率与畸胎率,并对出生小鼠(1w)的体重和性别比例进行分析。结果与对照组相比,灌注DEHP+DBP后,孕5.5天的胚胎早期植入率显著下降(77.5%Vs 62.5%,P<0.05);孕9.5天的畸胎率则显著上升(10.5%Vs 40.6%),差异具有统计学意义(P<0.01);出生小鼠的性别比例未发生变化,1周龄体重无统计学差异(3.74±0.31 Vs 3.72±0.33,P>0.05)。结论孕期邻苯二甲酸酯暴露会导致胚胎植入率下降、畸胎率升高,但对出生小鼠性别和体重无显著影响。
Objective To study the correlation between exposure to phthalates and embryonic development in mice during pregnancy. Methods Sixty female mice were randomly divided into six groups. Conclusion Exposure to phthalate during pregnancy may lead to decreased embryo implantation rate and teratogenic rate, but no significant effect on the sex and weight of the born mice.
