对健康献髓者在正常状态下和应用G-CSF时骨髓样本进行分析,以观察短期应用G-CSF对骨髓中总CD34~细胞数和早期CD34~+细胞亚群的作用。10名健康献髓者,中位年龄48岁(20~57岁),G-CSF皮下注射,10tμg/kg·d(7例)或10μg/kg·12h(3例)。分别在第1天(用G-CSF前),第3天(用G-CSF第2天),第5天(用G-CSF第4天)在局麻下从胸骨采骨髓1~1.5ml;在0,3,5天分别从静脉抽取外周血样。用Sysmex F-800计数骨髓和外周血细胞数,在FACS can上检测外周血和骨髓CD34~+细胞亚群。
Bone marrow samples from healthy volunteers under normal conditions and with G-CSF were analyzed to observe the effect of short-term administration of G-CSF on total CD34 ~ + cells and early CD34 ~ + subsets in bone marrow. Ten healthy donors were enrolled. The median age was 48 years old (20-57 years). G-CSF was injected subcutaneously at 10 μg / kg · d (n = 7) or 10 μg / kg · 12 h (n = 3). The bone marrow was collected from the sternum at 1 to 1.5 ml on the first day (before G-CSF administration), on the third day (on the second day of G-CSF administration), and on the fifth day (on the fourth day of G-CSF administration) Peripheral blood samples were taken from the veins on days 0, 3 and 5, respectively. The number of bone marrow and peripheral blood cells was counted by Sysmex F-800, and the peripheral blood and bone marrow CD34 + cell subpopulations were detected by FACScan.
