本研究旨在探讨激动素对D-半乳糖(D-gal)致衰老大鼠免疫功能的保护作用及其对脾淋巴细胞体外增殖的影响。50只SD大鼠随机分为5组,分别为青年对照组、衰老模型组、激动素低剂量组、激动素中剂量组和激动素高剂量组。除青年对照组外,其它组大鼠颈背部每天皮下注射125 mg/kg D-gal,持续45 d,制造衰老模型。从第11天开始,激动素低、中、高剂量组分别每天用5、10、20 mg/kg激动素溶液灌胃给药。检测血清IgG、IgA、IgM含量,取脾组织制备脾淋巴细胞悬液,检测体外脾淋巴细胞刺激指数(stimulation index,SI)、细胞凋亡率和细胞增殖指数(proliferation index,PI)等。结果显示,衰老模型组三种免疫球蛋白含量、SI和PI都较青年对照组降低,细胞凋亡率显著升高。激动素处理组血清免疫球蛋白含量、PI、SI较模型组升高,细胞凋亡较模型组降低,其中高剂量组这几项指标与青年对照组比较差异不显著。以上结果提示,激动素能有效抑制脾淋巴细胞的凋亡,刺激细胞增殖,从而提高免疫力,延缓机体的衰老。
This study aimed to investigate the protective effect of kinetin on immune function induced by D-gal in aging rats and its effect on the proliferation of splenic lymphocytes in vitro. Fifty Sprague-Dawley rats were randomly divided into five groups: young control group, aging model group, low-dose kinetin group, middle-dose kinetin group and high-dose kinetin group. Except for the youth control group, the rats in the other groups were subcutaneously injected with 125 mg / kg D-gal on the back of the neck for 45 days to produce the aging model. Starting on day 11, low, middle and high doses of kinetin were administered orally daily with 5, 10 and 20 mg / kg of kinetin solution. Serum IgG, IgA, IgM levels were measured. Splenic lymphocyte suspension was prepared from spleen tissue, spleen lymphocyte stimulation index (SI), apoptosis rate and proliferation index (PI) were detected in vitro. The results showed that the three immunoglobulin contents, SI and PI in the aging model group were lower than those in the youth control group, and the apoptosis rate was significantly increased. The content of serum immunoglobulin, PI and SI in the kinetin-treated group were higher than those in the untreated group, and the apoptosis was lower than that in the untreated group. There was no significant difference between the high-dose group and the young control group. The above results suggest that kinetin can effectively inhibit spleen lymphocyte apoptosis, stimulate cell proliferation, thereby enhancing immunity, delaying the body’s aging.
